In dieser Dissertation geht es um die Phototransduktionskaskade, das biochemische Reaktionsnetzwerk, welches in den Stäbchen in der Netzhaut die Umwandlung eines Lichtreizes in ein elektrisches Signal vollzieht. In der Arbeit wird diese Signalkaskade numerisch modelliert und es werden Simulationen unter verschiedenen Bedingungen durchgeführt: verschiedene Lichtreize, von sättigend bis hin zu einzelnen Photonen, und unterschiedliche genetische Knockouts der Proteine der Kaskade. Die Ergebnisse der Simulationen werden mit elektrophysiologischen Messungen verglichen. Ein besonderer Fokus liegt auf Antworten auf einzelne Photonen, die eine außergewöhnlich niedrige Variabilität haben. Für die Simulation dieser Lichtantworten wird ein stochastisches Modell entwickelt. Die Simulationen und die experimentellen Messungen werden statistisch ausgewertet und die Ergebnisse werden verglichen und interpretiert.
This dissertation examines the phototransduction cascade, a biochemical reaction network responsible for the translation of light stimuli into electrical signals in rod cells in the retina. In this thesis, this signaling cascade is modelled numerically and simulations are carried out for different conditions: various light stimuli, ranging from saturating to single photons, and genetic knockouts of different proteins of the signaling cascade. The simulation results are compared to electrophysiological measurements. Special emphasis is put on single photon responses, which exhibit a surprisingly low variability. A stochastic model is developed for the simulation of these responses. The simulations and experimental measurements are statistically analyzed, and the results are compared and interpreted.
Die Arbeit besteht aus drei Teilen. Im ersten Teil wurden verschiedene Diaminoterephthalsäurederivate synthetisiert. Ein zweites Themengebiet umfasste die Synthese einer monoprenylierten Modellverbindung sowie die Darstellung eines farnesylierten Diphosphats mit einer endständigen Alkinylgruppe, welches in biologischen Studien zur Anbindung eines Proteins dienen sollte. Im dritten Teil wurden verschiedene S-farnesylierten Cysteinderivate synthetisiert, die ebenfalls in biologischen Studien zur Untersuchung verschiedener Proteinfunktionen genutzt werden sollte.
This thesis consists of three parts. In course of the first part of this thesis different diaminoterephthalic acid derivatives were synthesized. The second topic includes the preparation of a monoprenylated model compound as well as the synthesis of a farnesylated diphosphate with a terminal alkyne. The latter one was supposed to be used in biological studies to be bound to a protein. Within the third part different farnesylated cysteine derivatives were synthesized, which also should have been used in biological studies to examine different protein functions.
