Die negativen Effekte von oxidativem Stress auf die Cochlea-Strukturen gelten als potentieller Faktor für sensorineuralen Hörverlust. Diese Arbeit untersucht, wie sich die fehlende Funktion der Glutathionperoxidase 1 (Gpx1), ein wichtiges antioxidatives Enzym, auf das Hören von Mäusen vor und nach Lärmbelastung auswirkt. Gpx1 KO Mäuse hatten eine verminderte Hörleistung, die nach Lärmbelastung im mittleren Alter nicht weiter abnahm. Da berichtet wurde, dass junge Gpx1 KO Mäuse lärmbedingten Hörverlust aufweisen, legt diese Arbeit nahe, dass die Ribbon-Entwicklung entscheidend für lärmbedingten Hörverlust sein könnte. Die Effekte von Gpx1 KO auf das Hören wurden durch die Expression der alternativen Oxidase (AOX) teilweise wiederhergestellt, jedoch waren Mäuse mit AOX-Expression anfälliger für Lärm. Die Ergebnisse deuten darauf hin, dass die ATP-Versorgung während der Lärmexposition für Hörfaserüberleben wichtiger ist, als das Risiko durch freie Sauerstoffradikale beschädigt zu werden.
Harmful impact of oxidative stress on cochlear structures is proposed as a contributing factor to sensorineural hearing loss. This thesis investigated how the missing function of Glutathione Peroxidase 1 (Gpx1), an major antioxidant enzyme counteracting oxidative stress, affects the hearing performance of mice before and after noise exposure (NE). Gpx1 KO mice had a reduced hearing performance. The hearing performance of middle-aged Gpx1 KO mice with NE did not decline further. As young Gpx1 KO mice were reported to show NIHL, this thesis suggests a critical role of ribbon maturity during NE for NIHL. Effects of Gpx1 KO on the hearing performance were partially rescued by supplementary expression of alternative oxidase (AOX). Surprisingly, mice with AOX expression were more susceptible to NE than control mice. The results suggested, that ATP supply during NE may be more crucial for auditory-nerve fiber survival than the risk of being damaged by increased reactive oxygen species levels.

Studien an Tieren und Menschen legen nahe, dass die zeitliche Wahrnehmung durch Alterung und Synaptopathie negativ beeinflusst wird. Das Ziel dieser Arbeit war es, einen solchen Vergleich durchzuführen mit Schwerpunkt auf Alterung und Synaptopathie bei der mongolischen Wüstenrennmaus.In der ersten Studie wurde die zeitliche Wahrnehmung und ihre Beziehung zur neuronalen Repräsentation untersucht. Die Ergebnisse zeigten, dass zeitliche Merkmale in der Hörbahn der Wüstenrennmaus bis zum Colliculus inferior erhalten sind. In der zweiten Studie wurden die Auswirkungen des Alters auf die zeitliche Wahrnehmung der zeitlichen Feinstruktur untersucht. Die Ergebnisse zeigten, dass alte Wüstenrennmäuse f0 bis zu 200 Hz diskriminieren können. In der dritten Studie wurden die Auswirkungen von Synaptopathie und Alterung auf die zeitliche Wahrnehmung untersucht. Die Ergebnisse deuten an, dass der Verlust von Fasern mit geringer spontaner Rate die zeitliche Wahrnehmung nicht beeinträchtigt.
Animal and human studies suggested that temporal perception is negatively affected by aging and synaptopathy. The aim of the thesis was to conduct such comparison by investigating temporal perception with focus on aging and synaptopathy in the Mongolian gerbil. In the first study, temporal perception and its relation to neural representation was investigated. The findings showed temporal features are preserved to some extent in the gerbil's auditory pathway up to the inferior colliculus.In the second study, the effects of age on temporal perception based on temporal fine structure cues were investigated. The results showed old gerbils can discriminate f0 up to 200 Hz In the third study, the effect of experimentally induced synaptopathy and aging on temporal perception was investigated. No differences were observed within the groups of young gerbils including the ouabain treated groups, suggesting low-spontaneous rate fiber loss did not affect temporal perception.
